Circadian Disruption May Open a Timing Window for Cancer Metastasis

Circadian rhythms influence not only sleep and metabolism but also the ability of cancer cells to invade, survive in blood, and colonize distant organs. This review examines how the circadian clock shapes the tumor microenvironment (TME) across the metastatic cascade, from invasive-edge remodeling and intravasation to circulation, extravasation, and colonization. It highlights rhythmic control of extracellular matrix (ECM) turnover, cancer-associated fibroblasts (CAFs), tumor-associated macrophages (TAMs), immune suppression, angiogenesis, circulating tumor cells (CTCs), endothelial adhesion, exosomes, and cell-cycle progression. The authors argue that circadian disruption can create time-dependent vulnerabilities in metastatic niches, suggesting that chronotherapy--treatment timed to biological rhythms--could complement existing cancer therapies.
